چکیده

THE literature on bacterial metabolism contains references which suggest that the anaerobic dismutation of pyruvic acid which we have shown to occur in animal tissues (see the preceding paper) may also take place in certain bacteria. Barron et al. [1932; 1933; 1934; 1936], for instance, described experiments concerning the oxidation of pyruvic acid in Gonococcus and showed that there is a specific mechanism by which pyruvic acid is oxidized to form acetic acid and carbon dioxide. He assumed that this oxidation is directly effected by molecular oxygen and he named the enzymic system concerned "ac-ketonoxidase". His data, however, also allow the assumption that the oxidation of pyruvic acid is brought about by the same type of dismutation which occurs in animal tissues:

Studies on Biological Oxidations

Pyruvic acid is one of the most reactive substances among those of importance in cellular metabolism. When carbohydrate is split during fermentation, pyruvic acid is the main end-product of the resulting series of phosphorylated oxidation-reductions. It thus becomes the immediate source for the formation of both lactic acid and alcohol. Pyruvic acid is decarboxylated by yeast; it may react with...

The Mechanism of the Enzymatic Synthesis of Cocarboxylase*

We have previously shown that the phosphorylation of vitamin B1 to cocarboxylase by an atiozymase preparation is dependent upon the presence of hexosediphosphate and certain heat-stable factors contained in boiled tissue extracts (1). Our results further indicated that this phosphorylation was completely inhibited by 0.0025 to 0.005 M sodium iodoacetate, and that the addition of 0.04 M sodium f...

Change of the tautomeric preference for radical cation of pyruvic acid. DFT studies in the gas phase

Keto-enol tautomerism was investigated for ionized pyruvic acid using the DFT(B3LYP) method and the larger basis sets [6-31++G(d,p), 6-311++G(3df, 3pd) and aug-cc-pVDZ]. Change of the tautomeric preference was observed when going from the neutral to ionized tautomeric mixture. Ionization favors the enolization process (ketoenol) of pyruvic acid, whereas the ketonization (ketoenol) is preferred ...

Biosynthesis of dicarboxylic acids by carbon dioxide fixation. III. Enzymatic synthesis of iota-malic acid by reductive carboxylation of pyruvic acid.

A complete chemical balance which demonstrates the occurrence of Reaction 3 has now been obtained. There is no reaction in the absence of either TPN, .Zwischenferment, or “malic” enzyme. The rate of reaction is proportional to the concentration of “malic” enzyme and to the CO* tension. It is also possible to couple the isocitric dehydrogenase-oxalosuccinic carboxylase system (2) with “malic” en...

The Relation of Thiamine to Citric Acid Metabolism*

The discovery of the identity of thiamine pyrophosphate and cocarboxylase and the recognition of the ester as the predominant form of thiamine in yeast (1, 2) and animal tissues (3, 4) has been followed by several papers which have clearly demonstrated that cocarboxylase is essential for the oxidative metabolism of pyruvic acid (5-7). However, it has become clear that pyruvic acid is one of the...
